Multiplex ligation-dependent probe amplification (MLPA)
Multiplex ligation-dependent probe amplification (MLPA) is a multiplex PCR-based technique that detects changes in the copy number of genes. The P027 MLPA (MRC-Holland) assay is designed specifically to detect the common gross chromosomal abnormalities found in uveal melanoma of chromosomes 1, 3, 6 and 8. MLPA is used routinely for prognostic testing of uveal melanoma specimens not only for patients treated at the LOOC but also when requested by clinicians from other hospitals. Through our research we have shown that MLPA has increased sensitivity of detecting those patients at high risk of developing metastasis, compared to alternative methodologies such as fluorescence in situ hybridisation.
